It is the diagnostic method whiacch is used for detection of autoantibodies which are bound to patient’s tissue.

  • Frozen section of patient’s tissue is placed over a slide.
  • Tissue is incubated with fluorescein­conjugated goat antihuman antibodies.

  • Section is viewed under ultraviolet microscope.

  • Frozen section of monkey’s esophagus is placed over a slide and is incubated with patient’s serum.
  • Excess serum is washed off
  • Apply fluorescent conjugated anti-human Ig antibodies.
  • Wash of excess antibody.
  • Section is viewed under ultraviolet microscope.
